A mitochondrial protonophore uncoupler that dissipates the proton gradient to burn calories as heat, promoting fat loss without affecting body temperature or food intake.

Overview

BAM15 is a mitochondrial uncoupling agent that works by partially dissipating the proton gradient across the inner mitochondrial membrane. Normally, this gradient drives ATP synthesis, but when uncoupled, the energy is released as heat instead. This effectively increases the body's metabolic rate and caloric expenditure. Unlike older uncouplers like DNP (2,4-dinitrophenol), BAM15 is selective for mitochondria and does not uncouple the plasma membrane, making it significantly safer with a much wider therapeutic window.

In preclinical studies, BAM15 has demonstrated impressive fat loss results — reducing body fat mass by up to 20% without affecting lean mass, food intake, or core body temperature. It also improves insulin sensitivity, reduces hepatic steatosis (fatty liver), and decreases inflammation.

Mechanism of Action

Selectively inserts into the inner mitochondrial membrane and facilitates proton leak across the membrane, partially uncoupling oxidative phosphorylation from ATP synthesis. This increases substrate oxidation (fatty acids and glucose) to maintain the proton gradient, effectively burning more calories. Unlike DNP, BAM15 does not uncouple the plasma membrane proton gradient.
