Overview
Kisspeptin is a neuropeptide encoded by the KISS1 gene that serves as the master upstream regulator of the hypothalamic-pituitary-gonadal (HPG) axis. Kisspeptin neurons in the hypothalamus directly stimulate GnRH (gonadotropin-releasing hormone) neurons, which in turn trigger the release of LH and FSH from the pituitary, ultimately driving testosterone and estrogen production. Kisspeptin is essential for puberty onset, fertility, and maintaining reproductive function throughout life. In health optimization, kisspeptin is used to naturally stimulate the HPG axis, supporting endogenous hormone production without the suppressive effects of exogenous hormones.
It has shown particular promise in treating hypogonadotropic hypogonadism, improving fertility in both men and women, and enhancing sexual desire through both hormonal and direct brain effects on limbic regions associated with sexual arousal.
Mechanism of Action
Binds to the KISS1R (GPR54) receptor on GnRH neurons in the hypothalamus, triggering GnRH pulsatile release. GnRH stimulates LH and FSH secretion from the anterior pituitary. LH drives testosterone production in Leydig cells (men) and estrogen in theca cells (women). Also activates limbic brain regions involved in sexual arousal.
