Overview
Pregnenolone is synthesized from cholesterol and serves as the foundational precursor for the entire steroid hormone cascade — it is converted into progesterone, DHEA, cortisol, aldosterone, testosterone, and estrogen through various enzymatic pathways. Often called the "mother hormone" or "grandmother hormone," pregnenolone occupies the apex of the steroidogenic pathway. Beyond its role as a precursor, pregnenolone is a potent neurosteroid in its own right. It is synthesized locally in the brain and enhances memory consolidation, promotes neuroplasticity, and modulates NMDA and GABA receptors. Pregnenolone sulfate (PS) is particularly active in the CNS, where it enhances long-term potentiation and memory formation.
Clinical interest has grown around pregnenolone for cognitive decline, mood disorders, and as part of comprehensive hormone optimization. It declines with age similarly to DHEA, and supplementation may help restore the body's ability to produce downstream hormones where needed.
Mechanism of Action
Converted from cholesterol by CYP11A1 (P450scc) enzyme. Serves as substrate for CYP17A1 (to DHEA pathway) and 3β-HSD (to progesterone pathway). As a neurosteroid, pregnenolone sulfate positively modulates NMDA receptors, negatively modulates GABA-A receptors (promoting alertness), and activates TRPM3 calcium channels. Enhances microtubule polymerization and axonal transport.
