Overview

Melanotan 1 (Afamelanotide) is a synthetic analog of alpha-melanocyte-stimulating hormone (alpha-MSH) that activates melanocortin 1 receptors (MC1R) on melanocytes, stimulating melanin production and providing UV-independent tanning. Unlike Melanotan 2, MT-1 is highly selective for MC1R and does not significantly activate other melanocortin receptors, giving it a cleaner side effect profile without the sexual, appetite, or cardiovascular effects associated with MT-2. Afamelanotide has been FDA-approved (as Scenesse) for the treatment of erythropoietic protoporphyria (EPP), a rare genetic condition causing extreme photosensitivity. In health optimization, Melanotan 1 is used for its photoprotective properties — the increased melanin provides natural UV protection, potentially reducing skin cancer risk and sun damage.

Mechanism of Action

Selectively binds and activates melanocortin 1 receptor (MC1R) on melanocytes. MC1R activation increases cAMP, which activates MITF (microphthalmia-associated transcription factor), upregulating tyrosinase and other melanin synthesis enzymes. Promotes eumelanin (brown/black protective pigment) production over pheomelanin.
