Semax
Also known as: ACTH(4-10) analog · Heptapeptide
A synthetic heptapeptide analog of ACTH(4-10) that enhances cognitive function, memory, and neuroprotection through BDNF upregulation.
Overview
Semax is a synthetic heptapeptide derived from the adrenocorticotropic hormone (ACTH) fragment 4-10, with an added Pro-Gly-Pro sequence at the C-terminus for enhanced stability and potency. Developed at the Institute of Molecular Genetics of the Russian Academy of Sciences, Semax has been approved in Russia and several CIS countries as a prescription nootropic and neuroprotective agent since the 1990s. Its primary mechanism involves upregulation of brain-derived neurotrophic factor (BDNF) and its receptor TrkB in the hippocampus — the brain region critical for memory formation and learning. BDNF is often called "Miracle-Gro for the brain" because it promotes neuronal growth, survival, and synaptic plasticity.
Semax also modulates the expression of genes involved in immune function, vascular regulation, and neurotransmitter systems. Clinical studies in Russia have demonstrated its efficacy in improving cognitive function after stroke, enhancing memory and attention in healthy individuals, and providing neuroprotection during cerebral ischemia. It is typically administered intranasally for rapid brain delivery.
Mechanism of Action
Upregulates BDNF and TrkB expression in the hippocampus. Modulates melanocortin system signaling. Enhances dopaminergic and serotonergic neurotransmission. Increases expression of neurotrophins NGF and NT-3. Activates the MAPK/ERK signaling pathway for neuronal survival.

Known Interactions
The following interactions have been documented for Semax. Always consult a healthcare professional before combining compounds.
Synergistic (3)
Classic nootropic stack. Semax boosts BDNF and cognitive performance while Selank reduces anxiety through GABA modulation. Complementary cognitive + anxiolytic effects.
Semax enhances BDNF and neuroplasticity while Alpha GPC provides acetylcholine precursors. Together they support both structural brain health and neurotransmitter production.
Dihexa (HGF/c-Met pathway) + Semax (BDNF pathway) targets two different neurotrophic systems for comprehensive cognitive enhancement.
